体内血桨凝块扩散盒(PCDC)培养中可以生长多种不同成熟程度的粒系和红系造血祖细胞。作者观察表明,正常小鼠骨髓在PCDC培养系统中至少生长有两种不同类型的粒系祖细胞。作者使用体内PCDC技术,同时以体外琼脂CFU-C和外源性CFU-S技术为对照,结合细胞形态学观察,以及注射细胞毒剂后体外孵育测定处于细胞周期中的细胞比例、水渗透崩溶和速度沉降等处理方法,测定了生长于PCDC培养中小鼠粒系祖细胞的特性。
A variety of maturational erythroid and erythroid hematopoietic progenitor cells can be grown in vivo in a clot diffusion chamber (PCDC) culture. The authors observed that the normal mouse bone marrow PCDC culture system in the growth of at least two different types of myeloid progenitor cells. In vivo PCDC technique was used in this study. In vitro CFU-C and exogenous CFU-S were used as control. Combined with cell morphological observation and in vitro incubation with cytotoxic agent, the proportion of cells in the cell cycle was determined. And velocity sedimentation and other processing methods were measured in the growth of PCDC cultured mouse myeloid progenitor cells.
