根据HPV6、11、16、18、31、33型L1ORF高度保守区的基因序列设计合成了一对HPVL1区共有引物。该引物除与上述六型有很高的同源性外还可覆盖其它已知的20多型与女性肛生殖区感染有关的HPV。用该引物及PCR技术对一组计144例活检标本进行检测,结果显示:病理确诊的尖锐湿疣、子宫颈癌及外阴癌组织、正常宫颈标本中HPVDNA序列检出率分别为100%(55/55)、70%(28/40)和13.3%(2/15)。另外,34例临床诊断而未被病理证实的尖锐湿疣HPV-DNA亦呈阳性,说明在对这类病变进行病理诊断时有必要进行病原学检测。
A pair of HPVL1 consensus primers were designed and synthesized according to the gene sequences of L1ORF highly conserved regions of HPV6, 11, 16, 18, 31 and 33. This primer, in addition to its high homology to the six types described above, can cover other known HPV types associated with 20 female genital infections. A total of 144 biopsy specimens were detected by PCR and polymerase chain reaction (PCR). The results showed that the detection rates of HPVDNA in condyloma acuminatum, cervical cancer and vulva cancer tissues and normal cervical specimens were 100% (55% 55), 70% (28/40) and 13.3% (2/15). In addition, HPV-DNA of 34 cases of condylomata acuminatum who were not clinically confirmed by pathology was also positive, which indicated that it is necessary to conduct etiological detection in the pathological diagnosis of such lesions.
