目的制备苦参素磷脂复合物并对其体外抗乙肝病毒活性进行评价。方法利用四氢呋喃作为反应溶剂,苦参素及磷脂溶于溶媒中,真空除去有机溶剂得苦参素磷脂复合物。并以2.2.15 cell line为细胞模型,分别对苦参素磷脂复合物的细胞毒性值和乙型肝炎表面抗原,乙型肝炎e抗原,乙型肝炎病毒DNA的抑制率进行了研究。结果制得的苦参素磷脂复合物中苦参素的含量为24.86%。苦参素磷脂复合物的细胞毒性值为250μg·mL-1。与苦参素相比,苦参素磷脂复合物对2.2.15细胞的HBsAg,HBeAg,HBV-DNA值明显降低。结论苦参素磷脂复合物能够产生更有效的抗乙肝病毒活性。因此乙肝治疗中苦参素磷脂复合物有很强的抗病毒活性潜力。
Objective To prepare the kushenin phospholipid complex and evaluate its anti-hepatitis B virus activity in vitro. Methods: Tetrahydrofuran was used as the reaction solvent, oxymatrine and phospholipid were dissolved in the solvent, and the organic solvent was removed in vacuo to obtain the kushenin phospholipid complex. The cytotoxic value of kushenin phospholipid complex and the inhibition rate of hepatitis B surface antigen, hepatitis B e antigen and hepatitis B virus DNA were studied by cell line model of 2.2.15 cell line. As a result, the content of oxymatrine in the oxymatrine phospholipid complex was 24.86%. The cytotoxic value of the kushenin phospholipid complex was 250 μg · mL -1. Compared with oxymatrine, the ratio of HBsAg, HBeAg and HBV-DNA in kenaf phospholipid complex was significantly decreased in 2.2.15 cells. Conclusion Oxymatrine phospholipid complex can produce more effective anti-HBV activity. Therefore, hepatitis C treatment of oxymatrine phospholipid complex has a strong anti-viral activity potential.
