目的探讨不同pH条件对活化血小板膜上表达CD62P、CD63测定的影响。方法利用流式细胞术分别测定30例正常健康人和30例脑血管疾病患者,在三种不同pH条件(pH6.0、pH7.4、pH8.8)下血小板膜糖蛋白(CD62P、CD63)的表达量。结果正常人在三种pH条件下对CD62P、CD63表达差异无显著性意义,脑血管疾病组在pH7.4条件下CD62P、CD63表达最高与pH8.8条件比较差异非常显著,而pH6.0条件与pH8.8条件下测定结果无显著性差异。结论pH7.4条件测定活化血小板膜上表达CD62P和CD63,可提高脑血管疾病的阳性检出率。
Objective To investigate the effect of different pH conditions on the expression of CD62P and CD63 on activated platelet membrane. Methods Flow cytometry was used to detect the expression of platelet membrane glycoproteins (CD62P, CD63) in 30 normal healthy subjects and 30 patients with cerebrovascular disease under three different pH conditions (pH6.0, pH7.4, pH8.8) The amount of expression. Results There was no significant difference in CD62P and CD63 expression between normal controls and normal controls. The highest expression level of CD62P and CD63 in cerebrovascular disease group was significantly different from that of pH8.8 at pH7.4, while pH6.0 No significant difference with the measured results under pH8.8 conditions. Conclusion The expression of CD62P and CD63 on activated platelet membrane measured by pH7.4 can improve the positive detection rate of cerebrovascular disease.
