目的探讨CD2 8/CTLA 4,B7在小鼠实验性变态反应言神经炎 (EAN )中的变化规律及对发病的影响。方法于小鼠双后肢足垫皮下注射兔坐骨神经匀浆 ,建立EAN模型 ,用免疫组化观察外周淋巴细胞CD2 8,CTLA 4,B7 1和B7 2蛋白的表达 ;用RT PCR检测外周淋巴细胞B7 1和B7 2mRNA的表达。结果B7 1蛋白和mR NA及CTLA 4蛋白从潜伏期至发病高峰呈递增趋势 ,而B7 2蛋白和mRNA及CD2 8蛋白从潜伏期至发病初期明显增高 ,至发病高峰又下降。结论CD2 8/CTLA 4,B7与EAN的发病密切相关。B7 2可能与EAN的始发有关 ,而B7 1与EAN的进展及维持关系更密切。
Objective To investigate the changes of CD2 8 / CTLA 4 and B7 in mice with experimental allergic neuritis (EAN) and the impact on the pathogenesis. Methods: EAN model was established by subcutaneous injection of rabbit sciatic nerve homogenate from the hindlimb footpad in mice. The expression of CD28, CTLA 4, B7 1 and B7 2 in peripheral lymphocytes was detected by immunohistochemistry. Peripheral lymph nodes Cell B7 1 and B7 2 mRNA expression. Results B7 1 protein and mR NA and CTLA 4 protein increased gradually from the incubation period to the peak of incidence, while the levels of B7 2 protein and mRNA and CD28 protein increased significantly from the incubation period to the onset of disease, and then decreased to the peak of incidence. Conclusion CD2 8 / CTLA 4, B7 and EAN closely related to the incidence. B7 2 may be related to the onset of EAN, while B7 1 is more closely related to the progress and maintenance of EAN.
