目的建立HPLC法同时测定不同产地北沙参中3种香豆素的含量。方法色谱柱为PhenomenexC18(250mm×4.6mm,5μm),流动相为甲醇水(65∶35),流速为1.0ml/min,检测波长248nm。结果补骨脂素在0.043μg~1.72μg(r=0.9999),欧前胡素在0.033μg~1.32μg(r=0.9999),异欧前胡素在0.030μg~1.20μg(r=0.9998)内线性关系良好;补骨脂素、欧前胡素和异欧前胡素平均回收率分别为97.59%、98.23%、96.94%,RSD分别为1.16%、1.36%、0.98%。结论不同产地北沙参中3种香豆素含量存在明显差异,可为评价不同产地北沙参质量提供依据。
Objective To establish a HPLC method for the simultaneous determination of three coumarins in different parts of North China Sea cucumber. Method The chromatographic column was Phenomenex C18(250mm×4.6mm, 5μm). The mobile phase was methanol water (65∶35). The flow rate was 1.0ml/min. The detection wavelength was 248nm. Results Psoralen ranged from 0.043 μg to 1.72 μg (r=0.9999), imperatorin ranged from 0.033 μg to 1.32 μg (r=0.9999), and isoimperatorin ranged from 0.030 μg to 1.20 μg (r=0.9998). The linear relationship was good; the average recoveries of psoralen, imperatorin, and isoimperatorin were 97.59%, 98.23%, and 96.94%, respectively, with RSDs of 1.16%, 1.36%, and 0.98%, respectively. Conclusion There are significant differences in the contents of three coumarins in different parts of the origin of the northern sea ginseng, which can provide a basis for evaluating the quality of northern sea ginseng in different producing areas.
