目的研究表皮生长因子受体酪氨酸激酶抑制剂盐酸埃克替尼对人舌鳞状细胞癌Tca8113细胞凋亡及Bcl-2、Bax蛋白表达的影响,探讨其诱导肿瘤细胞凋亡的可能机制。方法分别应用0、10、20、40μmol.L-1盐酸埃克替尼处理体外培养的人舌鳞状细胞癌Tca8113细胞,分别作用24、48、72 h后,采用流式细胞仪测定细胞凋亡率,采用间接免疫荧光联合流式细胞技术及免疫细胞化学方法检测盐酸埃克替尼对人舌鳞癌Tca8113细胞Bcl-2、Bax蛋白表达的影响。结果盐酸埃克替尼作用于人舌鳞癌Tca8113细胞后Bcl-2蛋白表达降低,Bax蛋白表达增加;随盐酸埃克替尼浓度、作用时间的增加,人舌鳞癌Tca8113细胞凋亡率增加;人舌鳞癌Tca8113细胞凋亡率与盐酸埃克替尼浓度、作用时间成正相关。结论盐酸埃克替尼可降低Bcl-2蛋白表达,增加Bax蛋白表达,诱导人舌鳞癌Tca8113细胞凋亡。
Objective To investigate the effect of epicatechin kinase inhibitor Icitinib on the apoptosis of tongue squamous cell carcinoma Tca8113 cells and the expression of Bcl-2 and Bax proteins, and to explore the possible mechanism of its induction of tumor cell apoptosis . Methods Human tongue squamous cell carcinoma Tca8113 cells were treated with 0, 10, 20, 40 micromol.L-1 Icitonitine, respectively. After 24h, 48h and 72h respectively, the apoptosis of cells was detected by flow cytometry The apoptosis rate was detected by indirect immunofluorescence combined with flow cytometry and immunocytochemistry. The effect of icotinine on the expression of Bcl-2 and Bax protein in Tca8113 human tongue squamous cell carcinoma was detected by indirect immunofluorescence staining. Results The expression of Bcl-2 protein and the expression of Bax protein were increased after treated with imatinib hydrochloride in human tongue squamous cell carcinoma cell line Tca8113. The apoptosis rate of human tongue squamous carcinoma cell line Tca8113 increased with the increasing of the concentration of imatinib hydrochloride The apoptosis rate of Tca8113 in tongue squamous cell carcinoma was positively correlated with the concentration of icotinib and the action time. Conclusion Icitinib hydrochloride can decrease Bcl-2 protein expression, increase Bax protein expression, and induce apoptosis of human tongue squamous cell carcinoma Tca8113 cells.
