目的 探讨骨形态发生蛋白 (BMP)对骨骼肌卫星细胞粘附特性的影响。方法 体外获取与培养骨骼肌卫星细胞 ,分别用 0、5 0、10 0、5 0 0、10 0 0 μg/L的BMP诱导培养基诱导培养 48h。通过荧光法测定接种后 1h的粘附细胞率 ,利用放射免疫法测定细胞层粘连蛋白的含量。结果 BMP可促进骨骼肌卫星细胞的粘附 ,在 5 0 0 μg/L的浓度时粘附率最高 ,当BMP浓度进一步增加时 ,细胞粘附率不再增加 ;BMP诱导后可促进骨骼肌卫星细胞层粘连蛋白的表达 ,当BMP浓度为5 0 0 μg/L时这种作用最强。 结论 BMP可增强骨骼肌卫星细胞的粘附特性 ,其机制与层粘连蛋白的表达增高有关。
Objective To investigate the effect of bone morphogenetic protein (BMP) on the adhesion of skeletal muscle satellite cells. Methods Skeletal muscle satellite cells were obtained and cultured in vitro. The cells were cultured in BMP induction medium of 0, 50, 100, 100, 100 microg / L for 48 hours respectively. The rate of adherent cells 1 h after inoculation was measured by fluorescence method, and the content of laminin was determined by radioimmunoassay. Results BMP could promote the adhesion of skeletal muscle satellite cells, with the highest adhesion rate at the concentration of 500 μg / L. When the BMP concentration was further increased, the cell adhesion rate did not increase any more. BMP induced skeletal muscle satellite The expression of laminin was strongest when the BMP concentration was 500 μg / L. Conclusion BMP can enhance the adhesion of skeletal muscle satellite cells. The mechanism is related to the increase of laminin expression.
