目的:深化造血细胞辐射损伤机理的认识,并为造血因子的合理应用提供理论指导。方法:应用MTT法观察4种造血因子依赖细胞株于照射后不同时间对6种造血因子的增殖反应性变化。结果:造血因子依赖细胞株受照后对造血因子的增殖反应性降低,表现为细胞因子ED50值的增加及最大增殖幅度的降低,照射剂量越大,反应性降低越明显,恢复所需时间越长。结论:增殖反应性降低可能是造血因子临床应用时剂量偏大的原因之一,并提示在造血细胞辐射损伤中可能伴有造血因子受体信号转导的障碍。
Objective: To deepen the understanding of the mechanism of hematopoietic cell radiation injury and provide theoretical guidance for the rational application of hematopoietic factors. Methods: MTT assay was used to observe the proliferative reactivity of four hematopoietic factor-dependent cell lines to six hematopoietic factors at different times after irradiation. Results: The hematopoietic cell-dependent cell lines showed decreased proliferative response to hematopoietic factors after irradiation, showing an increase in ED50 and a decrease in maximum proliferative rate. The greater the dose, the more significant the decrease in reactivity, the longer the time required for recovery long. CONCLUSION: Decreased proliferative response may be one of the reasons for the large dose of hematopoietic factor in clinical application, suggesting that hematopoietic cell radiation injury may be associated with disorder of hematopoietic receptor signaling.
