目的研究白芍总苷(TGP)对内毒素性心肌炎大鼠心肌组织中炎症因子TNF-α、IL-1β、NF-κB表达的影响。方法通过尾静脉注入脂多糖(LPS)建立内毒素性心肌炎大鼠模型,分别以低、中、高剂量的TGP进行干预,光镜观察大鼠心肌组织的病理学变化,免疫组化检测心肌组织TNF-α、IL-1β、NF-κB的表达水平。结果光镜观察发现模型组大鼠心肌组织炎症反应明显,较多炎症细胞浸润;TGP中、高剂量组大鼠心肌组织炎症反应较轻,有少量炎症细胞浸润;低剂量组大鼠心肌组织炎症坏死程度介于模型组和中剂量组之间。免疫组化检测提示TGP中、高剂量组TNF-α、IL-1β、NF-κB的表达明显减少,与模型组比较均有统计学差异(均P<0 01);TGP低剂量组的TNF-α、IL-1β表达与模型组均无统计学差异(均P>0.05),而NF-κB的表达与模型组有统计学差异(P<0.05)。结论 TGP能有效抑制内毒素性心肌炎大鼠心肌组织炎症反应,减轻心肌损伤,在中、高剂量时效果明显。
Objective To study the effects of total glucosides of paeony (TGP) on the expression of inflammatory cytokines TNF-α, IL-1β and NF-κB in myocardium of endotoxin-induced myocarditis in rats. Methods Lipopolysaccharide (LPS) was injected into the caudal vein to establish a rat model of endotoxic myocarditis. The rats were treated with low, medium and high doses of TGP respectively. The pathological changes of myocardial tissue were observed with light microscope. The myocardial tissue TNF-α, IL-1β, NF-κB expression levels. Results In the light microscope, the inflammatory reaction of myocardium in model group was obvious and the infiltration of inflammatory cells was more. TGP medium and high dose group had less inflammatory reaction in myocardium and infiltration of inflammatory cells in a small amount; myocardial inflammation in low dose group Necrosis between the model group and the middle dose group. Immunohistochemistry showed that the expression of TNF-α, IL-1β and NF-κB in TGP medium and high dose groups were significantly decreased compared with model group (all P <0.01) There was no significant difference between the model group and the model group (all P> 0.05), while there was a significant difference between the model group and the expression of NF-κB (P <0.05). Conclusion TGP can effectively inhibit myocardial tissue inflammatory response in endotoxic myocarditis rats and relieve myocardial injury, with obvious effect at medium and high doses.
