目的:观察平肝潜阳方对肾血管性高血压大鼠脑血管内皮P-选择素(CD62p)和细胞间黏附分子-1(ICAM-1)表达的影响。方法:将50只SD大鼠随机分为正常对照组10只,肾血管性高血压大鼠40只,肾血管性高血压大鼠随机分为4组:模型组、平肝潜阳法组、阿司匹林组、平肝潜阳法加阿司匹林组,每组10只,从造模后12周连续给药4周,造模16周后每组取5只视交叉平面连续切片,每8张取1张切片作原位杂交检测脑血管内皮的ICAM-1、CD62p的mRNA水平,剩余5只取视交叉平面连续切片,每8张取1张切片作免疫组化。结果:平肝潜阳法组或阿司匹林组治疗后脑血管内皮的CD62pI、CAM-1原位杂交阳性信号和免疫组化的阳性信号较模型组明显减弱(P<0.05);平肝潜阳法加阿司匹林组CD62pI、CAM-1原位杂交阳性信号和免疫组化的阳性信号较阿司匹林组或平肝潜阳法组亦均有明显减弱(P<0.05)。结论:肾血管性高血压大鼠术后第16周末脑血管内皮CD62pI、CAM-1表达水平明显增高;平肝潜阳法有明显抑制脑血管内皮CD62pI、CAM-1表达的作用,平肝潜阳法与阿司匹林合用治疗效果更显著。
Objective: To observe the effect of Pinggan Qianyang Recipe on the expression of P-selectin (CD62p) and intercellular adhesion molecule-1 (ICAM-1) in cerebral vascular endothelium in rats with renovascular hypertension. METHODS: Fifty SD rats were randomly divided into normal control group (n=10) and renovascular hypertensive rats (n=40). Renal vascular hypertensive rats were randomly divided into 4 groups: model group, Pinggan Qianyang group, The aspirin group and the Pinggan Qianyang plus Aspirin group consisted of 10 rats in each group and were continuously administered for 4 weeks from the 12th week after the model was established. After the 16th week of modeling, 5 ocular chiasmatic planes were serially sectioned, and every 8 were taken 1 The mRNA levels of ICAM-1 and CD62p in the cerebral vascular endothelium were detected by in situ hybridization in slices. The remaining 5 rabbits were consecutively sectioned at the chiasmatic plane and one of the 8 sections was used for immunohistochemistry. Results: Positive signals of in situ hybridization for CD62pI and CAM-1 and immunohistochemistry in the Pinggan Qianyang group or aspirin group were significantly lower than those in the model group after treatment (P<0.05); The positive signals of positive signals of in situ hybridization for CD62pI and CAM-1 in the aspirin group and immunohistochemistry were significantly lower than those in the aspirin group or the Pinggan Qianyang group (P<0.05). Conclusion: The expression of CD62pI and CAM-1 in the brain vascular endothelium of the renovascular hypertensive rats increased significantly at the end of the 16th week after operation. The Pinggan Qianyang method can significantly inhibit the expression of CD62pI and CAM-1 in the cerebral vascular endothelium. The combination of Yangfa and aspirin has a more significant effect.
