目的采用Q-TRAP LC-MS/MS测定桑杏汤水提液中活性成分绿原酸、苦杏仁苷、芦丁、贝母素甲和贝母素乙的含量。方法采用Supelco Discovery C-18(3μm,2.1 mm×150 mm)色谱柱,以乙腈和水(含体积比0.1%甲酸)为流动相,梯度洗脱,流速为0.25 ml/min,AB Sciex 4000 Q-TRAP型质谱仪多重反应监测(MRM)扫描模式对桑杏方水提液样品进行检测。结果桑杏汤水提液中绿原酸、苦杏仁苷、芦丁、贝母素甲和贝母素乙的含量分别为8.00,53.07,0.84,1.09μg/ml和0.67μg/ml。结论该方法简便、准确、重现性好,可用于对桑杏汤水提液中绿原酸、苦杏仁苷、芦丁、贝母素甲和贝母素乙这5种成分进行定量分析。
OBJECTIVE To determine the content of chlorogenic acid, amygdalin, rutin, peachin A and peiminine B in the aqueous extract of Sang Xing Tang by Q-TRAP LC-MS / MS. Methods A gradient elution was performed on a Supelco Discovery C-18 column (3 μm, 2.1 mm × 150 mm) using a mobile phase of acetonitrile and water (0.1% formic acid) with a flow rate of 0.25 ml / min and AB Sciex 4000 Q -TRAP mass spectrometer multiple reaction monitoring (MRM) scanning mode of Sang Xing Fang aqueous extract samples were tested. Results The contents of chlorogenic acid, amygdalin, rutin, peachin and peiminine in the extract of Sang Xingtang were 8.00, 53.07, 0.84, 1. 09μg / ml and 0.67μg / ml, respectively. Conclusion The method is simple, accurate and reproducible. It can be used to quantitatively analyze the five components of chlorogenic acid, amygdalin, rutin, peachin A and peiminine B in the extract of Sang Xing Tang.
