目的探讨DARPP-32在大鼠全脑的表达分布特点。方法应用免疫组织化学染色技术对大鼠脑内DARPP-32的表达分布进行观察。结果免疫组织化学染色结果显示,强阳性的DARPP-32染色大部分分布于基底节区和前嗅皮质区,主要分布在伏隔核、尾壳核及杏仁核复合体的神经元胞体内,以及苍白球、腹侧苍白球、脚间核及黑质网状部的神经纤维中;中~弱阳性DARPP-32免疫染色见于中脑红核、隔伞核、小脑的普肯耶神经元、内侧缰核、皮层及海马等区域;阳性DARPP-32主要定位于神经元的胞质、胞核和胞膜上。结论 DARPP-32在大鼠全脑广泛表达,但主要分布于多巴胺能神经元密集投射的脑区,这些区域的神经元富含多巴胺D1受体。DARPP-32可能在多巴胺介导的神经功能中扮演着重要角色。
Objective To investigate the distribution of DARPP-32 in rat brain. Methods The distribution of DARPP-32 in rat brain was observed by immunohistochemical staining. Results Immunohistochemical staining showed that most of the strongly positive DARPP-32 stains distributed in the basal ganglia and the pre-olfactory cortex, mainly in the neuronal soma of the nucleus accumbens, caudate putamen and amygdala Globus pallidus, ventral globus pallidus, nucleus of foot and nigral reticular nerve fibers; moderate ~ weak positive DARPP-32 immunostaining found in the midbrain red nucleus, septum nucleus, cerebellum Puconjeyer neurons, medial Halotus, cortex and hippocampus and other regions; positive DARPP-32 is mainly located in the neurons of the cytoplasm, nucleus and membrane. Conclusion DARPP-32 is widely expressed in the whole brain of rats, but mainly distributed in the brain areas where dopaminergic neurons are densely populated. The neurons in these areas are rich in dopamine D1 receptors. DARPP-32 may play an important role in dopamine-mediated neurological function.
