目的为满足新型战场止血需要,本研究研发了高岭土竹纤维止血纱布,观察其止血功能,并对其主要成分高岭土进行生物安全性评价。方法制备4种不同浓度的高岭土竹纤维止血纱布(0%,0.1%,0.5%,1%高岭土),建立兔急性出血动物模型;测定10 min各组止血量;流式细胞术(flow cytometry,FCM)检测各组外周血中血小板活化水平;HE染色观察受压组织形态学变化及纱布颗粒脱落物数量;以人血管内皮细胞(human vascular endothelial cells,HUVEC)细胞株为研究模型,观察高岭土的细胞毒性。结果与单纯竹纤维止血纱布组比较,0.5%,1%高岭土组10 min出血量显著减少;FCM检测发现,高岭土纱布能够更好的激活外周血中血小板的活化功能,其活化程度与高岭土呈现浓度依赖性;病理切片显示,各组纱布按压止血后,创面未发现纱布颗粒脱落物,但1%高岭土组出现了部分炎性细胞浸润。体外实验证实,高岭土具有良好的生物相容性,HUVEC均能在高岭土表面生长,并不会诱导细胞凋亡。结论高岭土竹纤维止血纱布具有显著的止血效果,且高岭土生物安全性好,其中含0.5%高岭土的竹纤维止血纱布为最理想的纱布。
Objective To meet the needs of new battlefield hemostasis, this study developed a kaolin bamboo fiber hemostatic gauze to observe its hemostatic function, and its main component of the bio-safety evaluation of kaolin. Methods Four kinds of kaolinite bamboo fiber hemostatic gauze (0%, 0.1%, 0.5%, 1% kaolin) were prepared at different concentrations. The animal models of acute hemorrhage in rabbits were established. The hemostatic volume of each group was measured at 10 min. Flow cytometry FCM was used to detect the level of platelet activation in peripheral blood of each group. The morphological changes of compressed tissues and the amount of gauze particles shedding were observed by HE staining. The cell population of human vascular endothelial cells (HUVEC) Cytotoxicity. Results Compared with the pure bamboo fiber hemostatic gauze group, the amount of bleeding in 0.5%, 1% kaolin group decreased significantly at 10 min. FCM showed that kaolin gauze could activate platelet activation in peripheral blood more effectively, Dependent. The pathological sections showed that gauze particles were not found on the wound surface after the hemostatic gauze of each group was pressed, but some inflammatory cells infiltrated in the group of 1% kaolin. In vitro experiments confirmed that kaolin has good biocompatibility, HUVEC can grow on the surface of kaolin, and does not induce apoptosis. Conclusion Kaolin bamboo fiber hemostatic gauze has a significant hemostatic effect, and the bio-safety of kaolin is good. Bamboo fiber hemostatic gauze containing 0.5% kaolin is the best gauze.
