目的分析缺氧诱导因子1α(HIF-1α)和核基质结合区结合蛋白质1(SATB-1)在肿瘤中的表达及与肿瘤分期的关系。方法收集手术切除的肿瘤标本120例,其中结直肠癌35例,乳腺癌28例,肾透明细胞癌23例,其他肿瘤34例。美国癌症联合委员会临床分期:Ⅰ期23例,Ⅱ期42例,Ⅲ期45例,Ⅳ期10例。同时取患者癌旁的正常组织标本40例作为对照。采用免疫组化SP法对所有标本进行染色处理,统计HIF-1α、SATB-1表达阳性率及免疫组化评分,分析其免疫组化评分和肿瘤分期的关系。结果肿瘤组织HIF-1α、SATB-1免疫组化评分高于癌旁正常组织[(9.5±2.0)分vs.(2.5±1.2)分、(10.3±2.5)分vs.(3.2±0.8)分](P<0.05)。肿瘤的分期越晚,其HIF-1α、SATB-1免疫组化评分越高(P<0.05)。结论肿瘤组织HIF-1α、SATB-1表达明显高于癌旁的正常组织。
Objective To analyze the expression of HIF-1α (HIF-1α) and nuclear matrix-binding domain-binding protein 1 (SATB-1) in tumor and its relationship with tumor staging. Methods Totally 120 resected tumor samples were collected, including 35 cases of colorectal cancer, 28 cases of breast cancer, 23 cases of clear cell renal cell carcinoma and 34 cases of other tumors. American Cancer Committee clinical stage: 23 cases of stage Ⅰ, 42 cases of stage Ⅱ, 45 cases of stage Ⅲ, 10 cases of stage Ⅳ. At the same time take the normal tissue specimens adjacent to the patient 40 cases as a control. All specimens were stained with immunohistochemical SP method. The positive rate of HIF-1α and SATB-1 expression and immunohistochemical score were calculated. The relationship between the immunohistochemical score and tumor stage was analyzed. Results The immunohistochemical scores of HIF-1α and SATB-1 in tumor tissues were significantly higher than those in normal tissues [(9.5 ± 2.0) vs. (2.5 ± 1.2) vs. (10.3 ± 2.5) vs. (3.2 ± 0.8) ] (P <0.05). The later the tumor staging, the higher the immunohistochemical score of HIF-1α and SATB-1 (P <0.05). Conclusions The expression of HIF-1α and SATB-1 in tumor tissues is significantly higher than that in adjacent normal tissues.
