目的探讨辛伐他汀对体外培养成骨细胞、破骨细胞的影响。方法分离SD大鼠成骨细胞、破骨细胞,体外培养时给予辛伐他汀刺激,观察药物对成骨细胞增殖率和碱性磷酸酶以及骨细胞骨吸收的影响。结果辛伐他汀10-6~10-9mol/L对碱性磷酸酶的分泌有明显促进作用,10-6~10-8mol/L的辛伐他汀既促进成骨细胞的增殖,又抑制破骨细胞的骨吸收。结论辛伐他汀在体外促进骨形成、抑制骨吸收,有望成为治疗骨质疏松的药物。
Objective To investigate the effect of simvastatin on osteoblasts and osteoclasts cultured in vitro. Methods SD rat osteoblasts and osteoclasts were isolated and stimulated with simvastatin in vitro. The effects of drugs on proliferation rate of osteoblasts, alkaline phosphatase and bone resorption were observed. Results Simvastatin 10-6 ~ 10-9mol / L significantly promoted the secretion of alkaline phosphatase, 10-6 ~ 10-8mol / L of simvastatin not only promote the proliferation of osteoblasts, but also inhibit osteoclast Bone resorption. Conclusion Simvastatin can promote bone formation and inhibit bone resorption in vitro and is expected to become a drug for the treatment of osteoporosis.
