本实验采用差速离心及不连续密度梯度离心法从人羊水中提取到肺表面活性物质,并分析其化学成分,测定其表面活性。肺表面活性物质的主要成分为磷脂,具有显著降低表面张力的作用,最小表面张力为7.2mN/m;当表面张力为10mN/m时压缩性为0.039m/mN,表面磷脂浓度为1.086nmol/Cm~2。为临床用以治疗新生儿呼吸窘迫综合征做准备,进行了无菌试验、热原试验、局部刺激试验等,均达到要求。
In this study, differential centrifugation and discontinuous density gradient centrifugation were used to extract pulmonary surfactant from human amniotic fluid, and its chemical composition was analyzed and its surface activity was measured. Pulmonary surfactant is the main component of phospholipids, with a significant role in reducing the surface tension, the minimum surface tension of 7.2mN / m; when the surface tension of 10mN / m compressibility of 0.039m / mN, the surface phospholipid concentration of 1.086nmol / Cm ~ 2. For the clinical treatment of neonatal respiratory distress syndrome preparation, conducted aseptic test, pyrogen test, local irritation test, have reached the requirements.
