左旋麻黄素半生物合成研究Ⅲ L-PAC(L-苯基乙酰基甲醇)的提取技术研究

本文对固相生物合成的L—PAC(左旋麻黄前体)提取条件进行了研究。结果表明,采用H型大孔吸附树脂从生物合成的反应产物中提取L—PAC最佳。当样品液PH为6左右,过柱流速为0.2—0.3BV/min、柱直径与树脂高度比为1.5:11,用质量分数为70%丙酮水溶液洗脱,该大孔吸附树脂对L—PAC的动态吸附量为61.69mg/g。洗脱的丙酮水溶液经减压浓缩,获油状L—PAC粗品,其质量浓度不低于O.24g/ml。 In this paper, the extraction conditions of solid-phase biosynthesis of L-PAC (Leprosy Ephedrine precursor) were studied. The results showed that L-PAC was best extracted from biosynthesis reaction products using H-type macroporous adsorption resin. When the sample liquid PH is about 6, the column flow rate is 0.2-0.3BV/min, the ratio of the column diameter to the resin height is 1.5:11, eluted with a mass fraction of 70% acetone solution, the macroporous adsorption resin to L-PAC The dynamic adsorption amount was 61.69 mg/g. The eluted acetone aqueous solution was concentrated under reduced pressure to obtain an oily crude L-PAC having a mass concentration of not less than 0.24 g/ml.
