目的:探讨慢性缺氧(chronic hypoxia,CH)及野百合碱(monocrotaline,MCT)诱导大鼠建立肺动脉高压(pulmonary hypertension,PH)形成差异及对肺动脉钙敏感受体(Ca2+-sensing receptor,Ca SR)表达影响。方法:分别通过CH及MCT诱导大鼠PH模型后检测实验组(缺氧3、7、14、21 d,MCT诱导3、7、14、21 d共8组)及对照(control,CON)组大鼠下列指标:右心导管法测大鼠肺动脉平均压力(mean pulmonary arterial pressure,m PAP),右心室肥大指数(right ventricular hypertrophy index,RVHI),取大鼠肺动脉以QPCR及Western blot方法检测Ca SR在肺动脉的表达情况情况。结果:CH组(CH 3、7、14、21 d)和MCT(3、7、14、21 d)大鼠m PAP、RVHI与CON组大鼠相比随干预时间延长(3、7、14、21 d)而升高,其中以CH 21d组和MCT 21 d组明显,而CH 21 d大鼠m PAP、RVHI均比MCT 21 d组低(P=0.000);Ca SR m RNA表达在CH组和MCT组随着干预时间延长而逐渐升高,其中在CH 21 d和MCT 21 d的时候最高,均高于CON组(P=0.000,P=0.000),而CH 21 d和MCT 21 d两者相比,MCT 21 d高于CH 21 d组,两者间有差异(P=0.000);在CON组大鼠,CH各组及MCT各组大鼠肺动脉中均检测到Ca SR蛋白的表达,并随着干预时间延长,Ca SR蛋白趋势逐渐升高,以CH 21 d和MCT 21 d明显,而MCT 21 d组高于CH 21 d组。结论:相同时间内MCT诱导大鼠的肺动脉压高于CH建立PH模型,Ca SR m RNA及蛋白表达水平与肺动脉压力呈正相关,Ca SR参与了PH的形成及发展。
Objective: To investigate the difference of pulmonary hypertension (PH) induced by chronic hypoxia (CH) and monocrotaline (MCT) in rats and its effect on the expression of Ca2 + -sensing receptor (Ca SR) ) Expression effect. Results: Compared with rats in CON group, the levels of m-PAP and RVHI in CH group (CH 3, 7, 14 and 21 days) and MCT (3, 7, 14 and 21 days) , 21 d). The levels of m PAP and RVHI in CH 21d group and MCT 21 d group were significantly lower than those in MCT 21 d group (P = 0.000). The expression of Ca SR m RNA in CH (P = 0.000, P = 0.000), while CH 21 d and MCT 21 d were the highest in CH 21 d and MCT 21 d MCT 21 d was higher than that of CH 21 d group (P = 0.000), and Ca SR protein was detected in the pulmonary arteries of CON rats, CH rats and MCT rats The expression of Ca SR protein gradually increased with the prolongation of intervention time, which was obvious on CH 21 d and MCT 21 d, but higher on MCT 21 d than CH 21 d. CONCLUSION: In the same time period, the pulmonary arterial pressure induced by MCT is higher than that of CH, and the PH model is established. The Ca SR m RNA and protein expression are positively correlated with pulmonary artery pressure. Ca SR is involved in the formation and development of PH.
