目的:探讨IL-10干预对肝纤维化大鼠表达血小板衍生生长因子-AA、BB(PDGF-AA、BB)的影响。方法:建立大鼠肝纤维化模型并行IL-10干预。分批从对照组(C组)和CCl_4诱导肝纤维化模型组(M组)及IL-10干预肝纤维化组(T组)中取肝脏组织,采用S-P免疫组织化学方法检测分析PDGF-AA、BB在肝纤维化进程中的表达状况和IL-10干预后不同组大鼠及同组不同阶段大鼠肝组织中的表达结果。结果:成功建立大鼠肝纤维化模型小组肝组织中PDGF-AA、BB阳性表达明显增强,且PDGF-BB阳性表达强度高于PDGF-AA。经Ridit分析,两者在C组与M组间比较阳性表达均有显著性差异(P<0.01);T组与M组间比较PDGF-AA无统计学差异,但T组的PDGF-BB较M组明显减弱;PDGF-BB在肝纤维化组与IL-10干预组不同阶段阳性表达差异均有显著性。结论:PDGF-BB阳性强度随肝纤维化进展表达增强,而抑制PDGF-BB的表达可能是外源性IL-10拮抗肝纤维化的机制之一。
Objective: To investigate the effect of interleukin-10 (IL-10) intervention on the expression of platelet-derived growth factor-AA and BB (PDGF-AA and BB) in rats with hepatic fibrosis. Methods: The rat model of hepatic fibrosis was established and IL-10 intervention was performed. The liver tissues were taken from the control group (C group) and the CCl 4 -induced liver fibrosis model group (M group) and the IL-10-induced liver fibrosis group (T group) in batches, and the expression of PDGF-AA was detected by SP immunohistochemistry , BB in the process of liver fibrosis and IL-10 intervention in different groups of rats and the same group of different stages of rat liver tissue expression results. Results: The positive expression of PDGF-AA and BB in liver tissue of rats successfully established hepatic fibrosis model group was significantly increased, and the positive expression of PDGF-BB was higher than that of PDGF-AA. Ridit analysis showed that there was a significant difference (P <0.01) between the two groups in the expression of PDGF-AA in group C and M (P <0.01). There was no significant difference in PDGF-AA between group T and group M M group was significantly reduced; PDGF-BB in liver fibrosis group and IL-10 intervention group at different stages of positive expression differences were significant. CONCLUSION: The positive expression of PDGF-BB increases with the progress of hepatic fibrosis, and the inhibition of the expression of PDGF-BB may be one of the mechanisms of exogenous IL-10 antagonizing hepatic fibrosis.
