为探讨bcr/abl融合基因对维持细胞生存方面的作用,通过脂质体介导的方法将表达bcr/abl融合区反义RNA片段的重组质粒导入K562和BV173细胞系,对细胞的超微结构、细胞大小的改变以及核DNA降解等进行观察。结果:表达bcr/abl反义RNA片段的细胞呈典型的细胞凋亡形态,细胞核DNA降解。证实bcr/abl反义RNA片段启动人Ph+白血病细胞系凋亡过程。提示bcr/abl融合基因是Ph+白血病细胞存活能力增强的重要因素,也为慢性髓系白血病的治疗指出了新的方向
To investigate the role of bcr / abl fusion gene in maintaining cell survival, a recombinant plasmid expressing the bcr / abl fusion region antisense RNA fragment was introduced into K562 and BV173 cell lines by liposome-mediated method. The ultrastructure of cell , Cell size changes and nuclear DNA degradation were observed. Results: The cells expressing bcr / abl antisense RNA showed typical apoptotic morphology and DNA degradation. Confirmed that the bcr / abl antisense RNA fragment initiates apoptosis in human Ph + leukemia cell lines. It is suggested that the bcr / abl fusion gene is an important factor for enhancing the viability of Ph + leukemia cells and also points out a new direction for the treatment of chronic myeloid leukemia
