目的:建立ITS区段的冬青属植物分子鉴别方法。方法:以15种冬青属植物为材料,采用离心柱型试剂盒提取总DNA,采用一对通用引物对ITS区段进行PCR扩增和测序;利用MEGA5.0软件分析,寻找差异位点,构建系统发育树。结果:测序后得到15种冬青属植物ITS比对序列为759 bp,种间存在216个差异位点。结论:ITS区段可以用于鉴别冬青属植物。
OBJECTIVE: To establish a molecular identification method of Ilex in ITS section. Methods: Fifteen kinds of holly plants were used as materials. Total DNA was extracted by centrifugal column kit. A pair of universal primers was used to amplify and sequence the ITS region. The MEGA5.0 software was used to find the difference sites and construct Phylogenetic tree. Results: The ITS sequence of 15 species of Ilex was 759 bp after sequencing, and there were 216 differences among species. Conclusion: The ITS segment can be used to identify Holly plants.
