目的:考察生物黏附剂卡波姆对淫羊藿总黄酮酶解效果的影响。方法:将淫羊藿总黄酮与卡波姆混合,加入蜗牛酶酶解,采用HPLC测定酶解液中朝藿定A、朝藿定B、朝藿定C和淫羊藿苷等主要黄酮成分及箭藿苷A、箭藿苷B、鼠李糖基淫羊藿次苷Ⅱ、宝藿苷Ⅰ和淫羊藿苷元等主要酶解产物的含量变化,并与未加入卡波姆时淫羊藿总黄酮的酶解过程比较。结果:在37℃,含1%卡波姆的缓冲溶液中,淫羊藿总黄酮中朝藿定A、朝藿定B、朝藿定C和淫羊藿苷的酶解速率变慢,约需4 h才能完全酶解转化成次级苷,所需时间为无卡波姆时的2倍;酶解液中宝藿苷Ⅰ质量浓度始终高于未加卡波姆时的,最高量48.43 mg·L-1,6 h时趋于稳定时12.99 mg·L-1,最高量和稳定量分别为未加卡波姆酶解时的2.2,7.2倍;2~6 h内箭藿苷A和鼠李糖基淫羊藿次苷Ⅱ质量浓度都略高于未加卡波姆时的;之后次级苷被继续水解生成苷元,且苷元生成速率基本相同,约7.60 mg·L-1·h-1。结论:卡波姆会减缓淫羊藿总黄酮的酶解速率,促进次级苷的生成,对苷元的生成基本无影响。

And rhamnosyl mycophenolate glycosides II were slightly higher than those without carbomer; the secondary glycosides were further hydrolyzed to produce aglycones, and the aglycone production rate was about 7.60 mg · L- 1 · h-1. Conclusion: Carbopol can slow down the enzymatic hydrolysis rate of total flavonoids of Epimedium, promote the formation of the secondary glycosides, and have no effect on the production of aglycones.
