目的观察新型含能材料3-硝基-1,2,4-三唑-5-酮(3-nitro-1,2,4-triagol-5-one,NTO)的亚慢性毒性。方法按照《化学品毒性鉴定技术规范》,选择健康成年SD大鼠96只,雌雄各半,随机分为对照组、NTO 250、500和1 000 mg/kg染毒组,每组24只。染毒组用NTO-植物油混悬液灌胃(10 ml/kg),对照组灌胃等量植物油,每日灌胃1次,每周5次,每周称重1次,连续灌胃90 d。观察大鼠一般情况、血常规、肝功能、脏器质量、大体解剖和病理组织学。结果中、高剂量组雄性大鼠体质量、肝脏系数和肾脏系数明显低于对照组(P<0.05)。低、中、高剂量组大鼠睾丸质量低于对照组。低、中、高剂量组雌性大鼠红细胞平均体积(MCV)升高,高剂量组雄性大鼠MCV升高,高剂量组雌性、雄性平均红细胞血红蛋白(MCH)升高。高剂量组大鼠肝脏、肾脏和睾丸病理组织学检查显示发生病变。结论 NTO染毒对SD大鼠生长发育、肝脏、肾脏和睾丸有一定损害作用。
Objective To observe the subchronic toxicity of 3-nitro-1,2,4-triagol-5-one (NTO), a new type of energetic material. Methods According to Technical Specifications of Toxicity Identification, 96 healthy adult SD rats were randomly divided into control group, NTO 250, 500 and 1 000 mg / kg, 24 rats in each group. The rats in the exposure group were orally administered with NTO-vegetable oil suspension (10 ml / kg). The control group was orally administered with the same amount of vegetable oil once a day for 5 times a week for 1 week. d. The rats were observed general, blood, liver function, organ quality, gross anatomy and histopathology. Results The body weight, liver coefficient and renal coefficient of male rats in high and medium dose groups were significantly lower than those in control group (P <0.05). The testicular weight of rats in low, medium and high dose groups was lower than that in control group. In the low, medium and high dose groups, the average volume of the red blood cells (MCV) in the female rats increased. The MCV in the high dose group increased, while that in the high dose group increased. High-dose group rats liver, kidney and testicular histopathology showed lesions. Conclusion NTO exposure has some damaging effects on the growth and development of liver, kidney and testis in SD rats.
