目的探讨2型糖尿病大鼠肾小管细胞聚核糖性死亡的形态学特点。方法 20只雄性SD大鼠分为正常组和2型糖尿病组,在光学显微镜和电子显微镜下观察两组大鼠肾小管细胞结构的变化,用免疫组织化学染色、免疫印迹方法检测多聚二磷酸腺苷核糖聚合酶-1(PARP-1)蛋白的表达。结果 2型糖尿病早期肾小管细胞内有大量糖原聚集,细胞肿胀,部分细胞坏死,主要为空泡样坏死。免疫组织化学染色显示,2型糖尿病肾小管上皮细胞PARP-1表达阳性。免疫印迹法显示2型糖尿病肾小管细胞中PARP-1蛋白表达增强。结论 2型糖尿病早期肾小管细胞发生了坏死,聚核糖性死亡(PARP-1介导的细胞死亡)参与了2型糖尿病早期肾小管细胞的损伤。
Objective To investigate the morphological characteristics of renal tubular polyglutamate death in type 2 diabetic rats. Methods Twenty male Sprague-Dawley rats were divided into normal group and type 2 diabetic rats. The changes of tubular cell structure of the two groups were observed under optical microscope and electron microscope. Immunohistochemical staining and immunoblotting were used to detect the expression of poly Adenosine ribose polymerase-1 (PARP-1) protein expression. Results In the early stage of type 2 diabetes, there was a large number of accumulation of glycogen in tubular cells, swelling of cells and partial necrosis of cells, mainly vacuole-like necrosis. Immunohistochemical staining showed that PARP-1 expression was positive in type 2 diabetic rat renal tubular epithelial cells. Immunoblotting showed enhanced expression of PARP-1 protein in tubular cells of type 2 diabetic rats. CONCLUSIONS: Early tubular cells of type 2 diabetes develop necrosis and polyribo-oligosaccharide death (PARP-1-mediated cell death) is involved in early tubular cell injury in type 2 diabetes.
