淋菌的检出和鉴别淋菌是肾形或咖啡豆形的双球菌,两凹面彼此相对,作横分裂增殖,其大小则依发育的阶段而不同,成熟的淋菌长约1.6微米,宽约0.8微米。在淋病的分泌物中,可见到淋菌集群在白血球内,当白血球破坏后可见到细胞外淋菌,保持集群状或散在性的双球菌。一般在急性期以在细胞内呈集群状为多,慢性期则以在细胞外呈散在性的双球菌为多。淋菌的染色是比较容易,如美蓝、甲基绿、龙胆紫等都可以,通常一般是用1%美蓝溶液来作单染色标本,这时淋菌和其他细菌染成深蓝色,细胞核染成蓝色,白血球的原浆和上皮细胞则染成淡蓝色。美蓝单染色标本制作法:(1)用白金耳取被检分泌物薄涂于载物玻片上。(2)干燥后通过火焰固定。(3)用1%美蓝溶液染色1—2分钟。(4)水洗,干燥,镜检(用油浸接物镜)。
