提出了真空微波预辐射辅助提取灵芝子实体多糖的新工艺,并利用响应面分析法(RSM)优化微波预处理工艺。根据正交旋转中心复合设计方案,采用三因素三水平的响应面分析法,在分析各因素显著性及其交互作用的基础上,得出灵芝子实体多糖提取的微波预处理最佳工艺条件:浸润时间60min,微波功率密度11.2W/g,微波辐射时间180s,灵芝子实体多糖的提取得率1.770%,并得到提取得率与工艺参数之间的数学模型。与传统热水提法比较,在料液比、提取温度相同的情况下,真空微波预处理法在提取时间缩短一半的基础上,提取得率提高47.75%。
A new technology of vacuum microwave pre-irradiation to extract polysaccharides from Ganoderma lucidum fruiting bodies was proposed, and the microwave pretreatment process was optimized by response surface analysis (RSM). According to the composite design scheme of orthogonal rotation center, three factors and three levels of response surface analysis method were used to analyze the significance of each factor and their interaction, and the optimum conditions of polysaccharide extraction from Ganoderma lucidum fruiting bodies were obtained: The wetting time was 60min, the microwave power density was 11.2W / g, the microwave irradiation time was 180s. The extraction rate of polysaccharide from Ganoderma lucidum fruiting body was 1.770%. The mathematical model was established between extraction yield and process parameters. Compared with the traditional hot water extraction method, the extraction rate increased by 47.75% on the basis of the shortened extraction time of vacuum microwave pretreatment method at the same feed-liquid ratio and extraction temperature.
