目的建立柱后光化学衍生-高效液相色谱法(HPLC)测定强化食品中叶酸的分析方法。方法采用SunFire C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-50 mmol/L磷酸二氢钾溶液(pH=5.0)为流动相梯度洗脱,叶酸在254 nm紫外光照射下进行柱后衍生,荧光检测器检测,激发和发射波长分别为344和428 nm。结果本方法的线性范围为0.01~2.0μg/ml,r=0.999 9;方法的定性检出限和定量检出限分别为0.01和0.03μg/g;加标回收率为90.0%~98.0%,RSD均小于5%。结论本方法简便、快速、准确、重现性好,适用于强化食品中叶酸含量的测定。
OBJECTIVE To establish a method for the determination of folic acid in fortified foods by photochemical derivatization with high performance liquid chromatography (HPLC). Methods The column was eluted with a SunFire C18 (4.6 mm × 250 mm, 5 μm) column and eluted with acetonitrile-50 mmol / L potassium dihydrogen phosphate solution (pH = 5.0) After derivatization, fluorescence detector detection, excitation and emission wavelengths were 344 and 428 nm. Results The linear range of the method was 0.01-2.0 μg / ml, r = 0.999 9. The detection limits of the method and the limits of detection were 0.01 and 0.03 μg / g, respectively. The recoveries of spiked samples ranged from 90.0% to 98.0% RSD less than 5%. Conclusion The method is simple, rapid, accurate and reproducible. It is suitable for the determination of folic acid in fortified foods.
