为了观察地黄多糖对创面愈合小鼠脾脏的影响,试验采用将60只昆明种小白鼠按随机数字表法分为3组:正常对照组、创伤模型组和地黄多糖治疗组;末次给药后1 h,取各组小鼠脾脏称重,计算脾指数;分离单细胞悬液碘化丙啶染色,采用FACSAria流式细胞仪进行检测,ModFit软件进行脾细胞增殖指数分析;并进一步检测脾脏淋巴细胞亚群的比例,60只小鼠,实验结果全部符合要求。结果表明:①与对照组比较,模型组脾指数明显降低;而治疗组脾指数升高。②与对照组比较,模型组脾细胞的增殖指数降低;治疗组可提高小鼠脾细胞的增殖指数。③与对照组比较,模型组CD3+CD4+与CD3+CD8+比值降低;治疗组CD3+CD4+与CD3+CD8+比值升高。说明地黄多糖通过提高创伤小鼠脾指数,促进脾淋巴细胞的增殖,纠正CD4+与CD8+比值,提高创伤小鼠脾脏免疫功能,进而促进创面愈合。
In order to observe the effect of Rehmannia glutinosa polysaccharide on the healing of wounds in spleen of mice, 60 Kunming mice were randomly divided into three groups according to random number table: normal control group, trauma model group and Rehmannia polysaccharide treatment group; h, spleen of each group were weighed and spleen index was calculated. Single-cell suspension propidium iodide staining was used to detect FACSAria flow cytometry, ModFit software was used to analyze spleen cell proliferation index, and spleen lymphocytes The proportion of subpopulations, 60 mice, the experimental results all meet the requirements. The results showed that: ①Compared with the control group, the spleen index of the model group decreased significantly; while the spleen index of the treatment group increased. ② Compared with the control group, the proliferation index of spleen cells in model group decreased; the treatment group could increase the proliferation index of spleen cells in mice. ③ Compared with the control group, the ratio of CD3 + CD4 + to CD3 + CD8 + in the model group decreased; the ratio of CD3 + CD4 + to CD3 + CD8 + increased in the treatment group. Rehmannia polysaccharide can improve spleen index of trauma mice, promote the proliferation of splenic lymphocytes, correct the ratio of CD4 + to CD8 +, and improve the spleen immune function of traumatized mice, which can promote wound healing.
