目的利用在大肠杆菌中表达的人骨形成蛋白-2(hBMP-2)成熟肽,制备兔抗hBMP-2成熟肽抗血清。方法将含有hBMP-2成熟肽基因的表达载体pDH2m,转化大肠杆菌,热诱导表达hBMP-2成熟肽,经纯化、复性后,再用其免疫兔子,制备兔抗hBMP-2成熟肽抗血清。并以Western-blot和免疫组化方法对抗血清进行鉴定。结果Western-blot结果显示在对应于hBMP-2成熟肽位置有阳性条带,免疫组化结果显示人软骨细胞胞浆阳性着色。结论hBMP-2成熟肽在大肠杆菌中获得高表达,并成功制备了兔抗hBMP-2成熟肽抗血清。
OBJECTIVE To prepare rabbit anti-hBMP-2 mature peptide antisera using mature human bone morphogenic protein-2 (hBMP-2) expressed in E. coli. Methods The hBMP-2 mature peptide gene expression vector pDH2m was transformed into E. coli and induced by heat induction to express hBMP-2 mature peptide. After purification and refolding, the rabbit anti-hBMP-2 mature peptide antiserum . The antiserum was identified by Western-blot and immunohistochemistry. Results Western-blot results showed that there was a positive band at the position corresponding to the mature peptide of hBMP-2, and immunohistochemistry results showed that human chondrocytes were cytoplasm positive staining. Conclusion The hBMP-2 mature peptide is highly expressed in Escherichia coli and the rabbit anti-hBMP-2 mature peptide antiserum was successfully prepared.
