目的观察唑来膦酸(ZOL)对血管内皮细胞CRL-1730放射敏感性的影响。方法血管内皮细胞CRL-1730经ZOL处理后,噻唑蓝比色法检测细胞活性,克隆形成实验检测细胞放射敏感性,小管形成实验检测内皮细胞血管形成能力,侵袭实验检测细胞转移能力。结果 ZOL处理呈剂量依赖性抑制CRL-1730细胞增殖能力;ZOL与电离辐射联用对CRL-1730细胞有明显的增敏效应,可抑制CRL-1730细胞克隆形成、小管形成及侵袭能力,同单纯ZOL用药组及单纯放射组比较差异显著(P<0.05)。结论 ZOL处理可有效提高血管内皮细胞CRL-1730的放疗敏感性。
Objective To observe the effect of zoledronic acid (ZOL) on the radiosensitivity of vascular endothelial cells (CRL-1730). Methods After treated with ZOL, the vascular endothelial cells CRL-1730 cells were assayed for cell viability by thiazolyl blue colorimetric assay, cell radiosensitivity by colony formation assay, angiogenesis ability of endothelial cells by capillary formation assay, and cell migration assay by invasive assay. Results ZOL treatment significantly inhibited the proliferation of CRL-1730 cells in a dose-dependent manner. ZOL combined with ionizing radiation had a significant sensitizing effect on CRL-1730 cells, which could inhibit the formation of colony, the formation of tubules and the invasiveness of CRL-1730 cells. There was significant difference between ZOL group and radiotherapy alone group (P <0.05). Conclusion ZOL treatment can effectively improve the radiosensitivity of CRL-1730 cells.
