目的 探讨光动力疗法 (PDT)对鼻咽癌细胞 (NPC)BaxmRNA表达和凋亡的影响。方法 体外培养的NPC细胞 ,分别在PDT作用 12、2 4、36和 48h后用电镜动态观察分析细胞凋亡现象和凋亡指数 ,同时采用逆转录 -聚合酶链反应技术 (RT PCR)检测BaxmRNA的表达情况。结果 PDT作用 12、2 4、36和 48h后NPC细胞凋亡指数分别为 6 .7%、71.6 %、86 .4%和 98.3 % ;同时BaxmRNA表达检测发现。各对照组NPC细胞仅可见BaxmRNA扩增产物 ,实验组NPC细胞BaxmRNA表达均呈阳性 ,随PDT作用时间延长而逐渐增强 ,两者结果呈现一致。结论 PDT体外可诱导鼻咽癌细胞凋亡 ,细胞凋亡指数上升与凋亡相关基因BaxmRNA表达上调有关。
Objective To investigate the effect of photodynamic therapy (PDT) on Bax mRNA expression and apoptosis in nasopharyngeal carcinoma (NPC) cells. Methods The apoptotic index and apoptotic index of NPC cells cultured in vitro were analyzed by electron microscope at 12,2,4,36 and 48 hours after exposure to PDT respectively. Meanwhile, Bax mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR) The expression of the situation. Results The apoptotic index of NPC cells after treated with PDT for 12, 24, 36, and 48 hours were 6.7%, 71.6%, 86.4% and 98.3%, respectively. Meanwhile, the expression of Bax mRNA was detected. The NPC cells in each control group showed only Bax mRNA amplification products. The expression of Bax mRNA in NPC cells in experimental group was positive, and gradually increased with the prolongation of PDT. The results of the two groups showed the same results. Conclusion PDT can induce the apoptosis of nasopharyngeal carcinoma cells in vitro. The increase of apoptosis index is related to the up-regulation of Bax mRNA expression.
