本文用ＰＥＧ６０００沉淀１０ｍｌ健康人混合精浆，粗提ＰＳＡ，再通过二次ＳｅｐｈａｏｌｅｘＧ－１５０柱层析，获得纯化的ＰＳＡ．经ＳＤＳ－ＰＡＧＥ分析为分子量约３４０００ｄｏｌｔｏｎ的单一条带。ＥＬＩＳＡ法测得含量为１．１０ｍｇ／ｍｌ，考马斯亮蓝法测其蛋白浓度为１．１４ｍｇ／ｍｌ．本法操作简便，所得产物经免抗人精浆血清证实达到免疫纯。
In this paper, PEG6000 precipitation 10ml healthy mixed seminal plasma, crude PSA, and then by two SepharosexG-150 column chromatography to obtain purified PSA. ELISA assay content of 1.10mg / ml, Coomassie brilliant blue method to measure the protein concentration of 1.14mg / ml. The method is simple and convenient, and the obtained product is proved to be immunopurified by anti-human seminal serum.
