目的探讨p38抑制剂SB203580(SB)对辐射诱导免疫细胞的影响。方法将C57BL/6小鼠分为对照组、照射组、SB组。对照组接受假照射,其余2组进行2Gy全身照射。SB 15 mg/kg腹腔注射,照射24 h后开始给药,隔日1次,共给药5次。SB末次给药24 h后处死小鼠,取外周血进行分型CD4、CD8、B220检测,取骨髓测定细胞内活性氧(ROS)。结果照射组外周血白细胞,B220比例明显下降,CD4细胞比例及骨髓细胞ROS水平显著增加,与对照组比较差异显著(P<0.05)。SB组外周血淋巴细胞比例升高,骨髓ROS水平下降,与照射组相比差异显著(P<0.05)。结论 p38抑制剂对辐射引起的免疫细胞分型变化有一定的影响。
Objective To investigate the effect of p38 inhibitor SB203580 (SB) on radiation-induced immune cells. Methods C57BL / 6 mice were divided into control group, irradiation group and SB group. The control group received fake irradiation and the remaining two groups received 2 Gy whole body irradiation. SB 15 mg / kg intraperitoneal injection, irradiation 24 hours after the start of administration, once every other day, administered a total of 5 times. Twenty-four hours after the last administration of SB, mice were sacrificed and peripheral blood was collected for the detection of CD4, CD8 and B220. Bone marrow was used to determine the intracellular reactive oxygen species (ROS). Results Compared with the control group, the ratio of B220 in the peripheral blood leukocytes in the irradiation group was significantly decreased, the proportion of CD4 cells and the level of ROS in the bone marrow cells were significantly increased (P <0.05). The proportion of peripheral blood lymphocytes in SB group increased, and the level of ROS in bone marrow decreased, which was significantly different from that in irradiated group (P <0.05). Conclusion The p38 inhibitor has a certain impact on the changes of immunocyte types induced by radiation.
