目的 :研究 p5 3和MGMT基因突变在急性白血病发病中的意义以及 p5 3基因突变与MGMT基因突变的关系。方法 :应用聚合酶链反应 单链构象多态性 (PCR SSCP )技术 ,对 36例急性非淋巴细胞白血病 (AN LL) ,2 6例急性淋巴细胞白血病 (ALL)和 2 3例健康对照组骨髓细胞的 p5 3和MGMT基因突变进行了分析。 结果 :p5 3基因突变率在ANLL中为 16 .7%、ALL中为 19.2 % ,MGMT基因突变率在ANLL中为 13.9%、ALL中为15 .4 % ;6 2例急性白血病中有 4例为 p5 3和MGMT 2个基因同时发生突变 ;正常对照组均未发生p5 3和MGMT基因突变。结论 :p5 3和MGMT基因突变在急性白血病发生与发展中起到一定作用 ,急性白血病中 p5 3基因突变和MGMT基因突变密切相关
Objective: To investigate the significance of p5 3 and MGMT mutation in the pathogenesis of acute leukemia and the relationship between p5 3 gene mutation and MGMT gene mutation. Methods: Thirty-six cases of acute non-lymphocytic leukemia (ANLL), 26 cases of acute lymphoblastic leukemia (ALL) and 23 healthy controls were studied by polymerase chain reaction single strand conformation polymorphism (PCR SSCP) Cell p5 3 and MGMT gene mutations were analyzed. Results: The mutation rate of p5 3 gene was 16.7% in ALL and 19.2% in ALL. The mutation rate of MGMT gene in ANLL was 13.9% in ANLL and 15.4% in ALL. Among 6 2 acute leukemias, 4 Mutations of p5 3 and MGMT genes were observed at the same time. No mutation of p5 3 or MGMT gene was observed in normal control group. Conclusion: The mutations of p5 3 and MGMT play a role in the development and progression of acute leukemia. The mutation of p5 3 gene and the mutation of MGMT gene in acute leukemia are closely related
