目的 采用原子力显微镜对奇异变形杆菌菌落边缘的细菌排列及菌间关系进行观察。方法 切取菌落边缘琼脂小块 ,将琼脂小块上的菌落轻轻印在云母片上 ,于原子力显微镜下观察。结果 奇异变形杆菌波动菌落边缘细菌成单层紧密排列 ,菌体周围有大量鞭毛存在并在菌体之间耦合成束 ,菌体表面见到密集的蛋白颗粒。结论 原子力显微术可在生理条件下原位观察活体细菌膜表面结构及菌体间相互关系 ,此方法操作简单 ,成像清晰且分辨率高
Objective To observe the arrangement of bacteria and the relationship between bacteria in the margins of Proteus mirabilis by atomic force microscopy. Methods The colony edge agar cuttings were cut and the colonies on the agar pellet were gently printed on mica plate and observed under atomic force microscope. Results The bacteria on the edge of the colonies were closely arranged in the monolayer. There were a large number of flagella surrounding the cells and the bundles of bacteria were coupled into bundles. The surface of the cells was densely packed with protein particles. Conclusion Atomic force microscopy can observe the surface structure of bacterial membranes in vivo and the relationship between the bacterial cells in situ under physiological conditions. This method is simple in operation, with clear imaging and high resolution
