[目的]探讨在CV-1细胞中肝细胞核因子-1α(HNF-1α)对小鼠Ⅰ型葡萄糖载体(mGLUT1)表达的影响.[方法]利用DNA克隆技术,将HNF-1αcDNA序列重组到表达载体pSV-sport上,以HNF-1α转染CV-1细胞,应用Northern blot方法测定HNF-1α对mGLUT1 mRNA表达的影响.[结果]HNF-1α可提高mGLUT1 mRNA表达水平及稳定性.[结论]HNF-1α可增强mGLUT1的表达,可能通过调节GLUT1的表达参与胰岛素对血糖的调节过程.
[Objective] To investigate the effect of HNF-1α on the expression of mGLUT1 in mouse CV-1 cells. [Methods] The HNF-1α cDNA sequence was recombined to express The effect of HNF-1α on the expression of mGLUT1 mRNA was detected by Northern blot. [Results] HNF-1α could enhance the expression level and stability of mGLUT1 mRNA in CV-1 cells. [Conclusion] HNF- ] HNF-1α can enhance the expression of mGLUT1, which may be involved in insulin regulation of blood glucose by regulating the expression of GLUT1.
