目的:探讨慢性复合式应激3周对雄性小鼠生育能力的影响以及中药的干预作用。方法:昆明种雄性小鼠随机分为正常组、应激组、应激加六味地黄丸组、应激加加味逍遥丸组。将心理应激和束缚、尾悬挂、站台、夹尾搏斗五种应激方法随机安排到3w内,每天一种应激方法,相邻两天不重复。自应激之日起开始喂药,观察行为;于末此应激后第二天,取血清、睾丸、精子进行各项分析。结果:五种应激方法组成的复合式应激方式能显著降低应激组动物的体重增长率、精子活率和精子大小,病理分析睾丸生精上皮的各项评价指标和管腔精子量明显降低。精子内PH值和血清激素FSH、T水平也显著降低。两种药物对精子大小、睾丸生精上皮的排列和凋亡评分、管腔精子量及精子内PH值等指标都有显著的疗效;而六味地黄丸对精子的活率、生精上皮的分化程度、生精细胞的凋亡及血清FSH水平的影响大于加味逍遥丸。结论:五种应激模式组成的慢性复合式应激能显著改变雄性小鼠与生育有关的生理特性,使用六味地黄丸和加味逍遥丸干预后,各项指标都有不同程度的改变,其中六味地黄丸对于部分指标的影响大于加味逍遥丸。
OBJECTIVE: To investigate the effect of chronic combined stress on the fertility of male mice and the intervention of traditional Chinese medicine for 3 weeks. Methods: Kunming male mice were randomly divided into normal group, stress group, stress plus Liuwei Dihuang Wan group, and stress plus Jiawei Xiaoyao pill group. Psychological stress and restraint, tail suspension, platform, and tail-fighting were randomly arranged within 3 days. One stress method per day was not repeated for two days. The drug was started on the day of stress and the behavior was observed. On the second day after the stress, serum, testis, and sperm were analyzed. RESULTS: The composite stress method consisting of five stress methods could significantly reduce the body weight gain rate, sperm viability and sperm size of the stressed animals. Pathological analysis of the testicular spermatogenic epithelial evaluation indicators and the amount of lumen sperm reduce. The sperm pH and serum hormone FSH, T levels were also significantly reduced. Both drugs had significant effects on sperm size, arrangement of spermatogenic epithelium and apoptosis scores, luminal sperm content, and pH value in sperm; while Liuwei Dihuang Pill had a high sperm viability and differentiation of spermatogenic epithelium. The degree of apoptosis, apoptosis of spermatogenic cells and serum FSH levels were greater than those of Modified Xiaoyao Pills. Conclusion: The chronic compound stress consisting of five stress modes can significantly change the physiological characteristics of male mice. After intervention with Liuwei Dihuang Pill and Jiawei Xiaoyao Pill, the indexes have changed to varying degrees, among which Liuwei Dihuang Pills have more influence on some indicators than Jiawei Xiaoyao Pills.
