中药党参的水提液经乙醇沉淀,酶法处理后,经Sevag法脱蛋白,对蒸馏水充分透析,经DEAE-SephadexA-50柱层析,得FP-l和FP-2两个多糖组分,二者产率比为l:3。FP-l和FP-2经醋酸纤维薄膜电泳,纸电泳,玻璃纤维纸高压电泳和凝胶柱层析都证明是一均一组分,经凝胶过滤法测得FP-l和FP-2的分子量分别为50万和2万左右。 FP-l和FP-2的旋光性均为正值,比旋度分别为210°和20.5°用Discher法测试表明,二者均含有糖醛酸。经纸层析,薄板单向层析,双向层析分析,FP-l含鼠李糖,甘露糖、半乳糖和葡萄糖醛酸。FP-2含木糖、半乳糖醛酸和一未知糖。红外光谱,H-核磁共振测定结果表明,FP-1糖苷健构型为β-型,FP-2为α-型。 FP-1和FP-2经高碘酸氧化及Smith降解,表明FP-l以1→3连接键存在,而FP-2除了以l→3为主要连接方式,尚有少量的l→6和1→4键连接的分枝链。
The aqueous extract of Radix Codonopsis Pilosulae was ethanol-precipitated, enzymatically treated, deproteinated by Sevag method, and dialyzed against distilled water. The DEAE-Sephadex A-50 column chromatography gave two polysaccharide fractions, FP-1 and FP-2. The yield ratio of the two is l:3. FP-l and FP-2 were analyzed by cellulose acetate membrane electrophoresis, paper electrophoresis, glass fiber paper high-pressure electrophoresis and gel column chromatography, and the FP-1 and FP-2 were measured by gel filtration method. The molecular weights are 500,000 and 20,000 respectively. The optical rotations of FP-1 and FP-2 were positive, and the specific rotations were 210° and 20.5°, respectively. The Discher test showed that both contained uronic acid. After paper chromatography, one-way chromatography of thin plates and two-way chromatography analysis, FP-l contained rhamnose, mannose, galactose, and glucuronic acid. FP-2 contains xylose, galacturonic acid and an unknown sugar. Infrared spectroscopy and H-NMR measurements showed that the FP-1 glycoside is β-type and FP-2 is α-type. FP-1 and FP-2 were subjected to periodic acid oxidation and Smith degradation, indicating that FP-I existed as a 1→3 linkage, while FP-2 contained a few l→6 and l→3 as the main linkage. 1→4 key connected branch chain.
