目的:分析猪囊尾蚴特异性抗原编码cDNA的阶段特异性表达机制。方法:用猪囊尾蚴特异性抗原编码cDNA(λcC28)及抗λcC28单表位抗体作探针,采用Northernblot、Southernblot和Westernblot分子杂交技术分析了λcC28编码cDNA在猪带绦虫各发育阶段的特异性表达。结果:猪囊尾蚴和成虫不同节片抗原蛋白具有阶段特异性,λcC28cDNA编码的28kDa抗原蛋白只在囊尾蚴阶段表达,成虫幼节、成节、孕节都不表达。结论:猪囊尾蚴阶段特异性抗原的编码基因存在于囊尾蚴阶段和成虫的幼节中,但仅有囊尾蚴阶段的mRNA表达28kDa抗原蛋白。
Objective: To analyze the stage-specific expression of cDNA encoding cysticercus cellulosae specific antigen. Methods: The specific cDNA of λcC28 encoding cDNA was amplified by cDNA microarray (λcC28) and anti-λcC28 single epitope antibody. Northern blot, Southern blot and Western blot were used to analyze the specific expression of λcC28 cDNA in various developmental stages of Taenia solium . Results: The antigen of different cytoplasm of Cysticercus cellulosae and adult was phase-specific. The 28kDa antigen protein encoded by λcC28cDNA was expressed only in cysticercus stage. CONCLUSION: The coding gene of stage-specific antigen of Cysticercus cellulosae exists in the cysticerci stage and in the adult larvae, but only the cysticercus stage mRNA expresses the 28 kDa antigen protein.
