目的研究心梗后心力衰竭(心衰)大鼠的海马区基因表达变化特征。方法采用结扎左冠状动脉前降支的心梗模型,利用基因表达谱芯片技术,研究大鼠心衰10 d和心衰60 d,海马区和皮质区的基因表达变化。结果心衰10 d大脑的基因变化明显比心衰60 d时活跃,心衰10 d时海马区的阳性差异基因为24条,而心衰60 d时为14条,主要涉及到的有能量代谢、神经递质代谢、神经鞘脂、信号转导、应激和免疫反应等方面作用的基因。其中,能量代谢类基因丙酮酸激酶3(NM_053297)和3-羟3-甲基辅酶A裂解酶(NM_024386)基因在心衰10 d时下调;神经递质代谢类基因烟碱型ε乙酰胆碱能受体(NM_017194)、alpha Endosulfine(NM_021842)和谷氨酸草酰乙酸转氨酶2(NM_013177)基因在心衰10 d时下调。结论心衰初期大脑海马区的能量代谢类基因和神经递质代谢类基因发生了变化。
Objective To study the gene expression changes of hippocampus in heart failure (CHF) rats after myocardial infarction. Methods Myocardial infarction model was established by ligation of the left anterior descending coronary artery. Gene expression profiling was used to study the gene expression changes in the hippocampus and cortex after 10 d of heart failure and 60 d of heart failure. Results The gene expression of brain in 10-day heart failure was significantly higher than that in 60-day heart failure. The positive differential gene in hippocampus was 24 at 10 days of heart failure, and 14 at 60 days of heart failure. The main energy metabolism , Neurotransmitter metabolism, sphingolipid, signal transduction, stress and immune response and other aspects of the gene. Among them, the genes of energy metabolism genes pyruvate kinase 3 (NM_053297) and 3-hydroxy-3-methyl-CoA lyase (NM_024386) were downregulated on the 10th day of heart failure; the neurotransmitter metabotropic genes nicotinic acetylcholine Body (NM_017194), alpha Endosulfine (NM_021842) and glutamate oxaloacetate aminotransferase 2 (NM_013177) genes were down-regulated at 10 days of heart failure. Conclusion The energy metabolism genes and neurotransmitter metabolism genes of hippocampus in the early stage of heart failure have changed.
