原癌基因经过突变,失去了正常的生长控制而成为癌基因,首先是细胞形态发生了改变。ras癌基因编码的蛋白能使细胞发生表型的改变ras基因的点突变往往是在P21蛋白第12位氨基酸和第61位氨基酸上的一个核苷酸发生改变。本文报道了将抗第12位氨基酸发生突变的v-Ki-ras癌基因蛋白的特异抗体,显微注射到ras蛋白转化的Ki-NRK细胞中,能使转化的细胞发生瞬间的逆转。 ras蛋白定位在细胞质膜中并和GTP·GDP结合,它的功能类似于腺苷酸环化酶体系的“G”调节蛋白,能影响cAMP的水平。

In this paper, the specific antibody of v-Ki-ras oncoprotein which is mutated to the amino acid at position 12 was microinjected into the ras protein-transformed Ki-NRK cells to reverse the transformed cells transiently. The ras protein, located in the plasma membrane and bound to GTP · GDP, functions similarly to the “G” regulatory protein of the adenylate cyclase system and affects cAMP levels.
