为研究冬凌草多糖的理化性质和质量,对冬凌草采用热水、稀碱提取,乙醇沉淀,等电点法脱蛋白后得到冬凌草粗多糖,经Q Sepharose 4 Fast Flow离子交换层析柱和Sephacryl S-100凝胶柱进一步纯化后得到4个纯度较高的组分STPS-Ⅰ、STPS-Ⅱ、JTPS-Ⅰ、JTPS-Ⅱ。对各组分的总糖含量、糖醛酸含量、硫酸根含量以及蛋白含量进行了测定,利用高效液相色谱法测定了单糖组成和分子量。结果显示冬凌草多糖不同组分的分子量相近,化学组成及单糖组成明显不同。
In order to study the physicochemical properties and quality of Rubescens polysaccharide, rubescens rubescens polysaccharides were obtained by hot water, dilute alkali extraction, ethanol precipitation, isoelectric point deproteinization and purified by Q Sepharose 4 Fast Flow ion exchange layer After purification by column and Sephacryl S-100 gel column, four components STPS-Ⅰ, STPS-Ⅱ, JTPS-Ⅰ and JTPS-Ⅱ were obtained. The total sugar content, uronic acid content, sulfate content and protein content of each component were determined. The composition and molecular weight of monosaccharide were determined by high performance liquid chromatography. The results showed that the molecular weights of different components of the polysaccharides of Rubestein were similar, but the chemical composition and the composition of monosaccharides were obviously different.
