近年来,由于半薄切片机的问世,为电镜工作者制作半薄切片进行定向选择,克服超薄切片的盲目性和电镜视野的局限性,创造了良好条件。但是,国内目前所使用的半薄切片机仍靠进口,价格昂贵。而定位部分的照明光源又不能达到满意效果,对定向选择带来一定困难,操作麻烦,花时间多。两年来,我室利用超薄切片机制作半薄切片,对心肌、肾等组织进行定向选择,省时省力,并获得了满意的效果。现将具体操作步骤介绍如下:
In recent years, due to the advent of semi-thin slicing machines, SEM workers make semi-thin slices for directional selection, overcome the blindness of ultra-thin slices and limitations of electron microscopy, and create favorable conditions. However, the semi-thin slicer currently used in China is still imported and expensive. The positioning part of the light source can not achieve satisfactory results, the orientation of choice to bring some difficulties, the operation of trouble, take more time. In the past two years, our laboratory made semi-thin slices by using ultra-thin slicer to make directional selection on tissues such as myocardium and kidney, saving time and labor and achieving satisfactory results. The specific steps are described below:
