参莲提取物对TNF-α刺激下单核细胞株THP-1功能的影响

目的:观察参莲(SL)提取物对炎症因子刺激下单核细胞功能的影响。方法:SL提取物预孵育,以TNF-α20μg.L-1为刺激因子,观察SL提取物对单核细胞株THP-1黏附、迁移、摄脂、分泌功能变化的影响。结果:SL1~3(0.5~2 g.L-1)可明显降低HUVEC和THP-1的黏附率,与模型组比较有显著性差异。SL1~5(0.125~2 g.L-1)均可显著减少THP-1迁移细胞数,与模型组比较有显著性差异。SL1~5(0.125~2 g.L-1)均可显著降低细胞内TC值。SL 1(2 g.L-1)可显著降低细胞上清中异常增高的IL-6值,同时升高IL-10的含量,其他剂量未见明显作用。结论:SL提取物对TNF-α刺激下单核细胞黏附、迁移、摄脂、分泌功能均有显著影响,其作用可能是SL提取物干预AS早期炎症反应的机制之一。 Objective: To observe the effects of the extract of Shen-Lin (SL) on the function of monocytes stimulated by inflammatory cytokines. Methods: The SL extract was pre-incubated and TNF-α (20μg.L-1) was used as a stimulant to observe the effect of SL extract on the adhesion, migration, lipofection and secretory function of monocytic cell line THP-1. Results: SL1 ~ 3 (0.5 ~ 2 g.L-1) could significantly reduce the adhesion rate of HUVEC and THP-1, which was significantly different from the model group. SL1 ~ 5 (0.125 ~ 2 g.L-1) can significantly reduce the number of THP-1 migrating cells, compared with the model group were significantly different. SL1 ~ 5 (0.125 ~ 2 g.L-1) can significantly reduce the intracellular TC value. SL 1 (2 g.L-1) significantly decreased the abnormally elevated IL-6 in the cell supernatant and increased the content of IL-10, while no significant effect was observed in other doses. CONCLUSION: SL extract has a significant effect on the adhesion, migration, lipofection and secretion of monocytes stimulated by TNF-α, which may be one of the mechanisms by which SL extract interferes with the early inflammatory response of AS.
