从胡萝卜胚cDNA文库中分离得到一条完整的cDNA片段,序列分析表明属于水通道家族新成员,并与烟草根特异性表达TobRB7基因高度同源,因此命名为DcRB7.Northern杂交分析表明DcRB7在根中特异性表达.利用反向PCR技术,分离获得了DcRB7基因上游约650bp的片段.将此片段与含有GUS报告基因表达载体进行重组构建后,以农杆菌介导转化烟草获得相应的转基因植株.在对转基因烟草的GUS表达特性进行了组织化学鉴定和荧光定量分析后发现,该调控区段能指导GUS基因在烟草根中优势表达,并表现出干旱诱导的特性.

It was found that the GUS gene was predominantly expressed in tobacco roots and showed drought-induced characteristics.
