本文报道制备固定化苯丙氨酸解氨酶(PAL)的初步结果。采用包埋和共价结合两种方法将PAL固定化。结果表明前者的偶联率高于后者。冷冻干燥,4°下贮存6个月后,活性存留分别为89%和76%,说明固定化后稳定性良好。包埋法采用不同T/C比(T为凝胶总浓度;C为交联百分比)的聚丙烯酰胺凝胶(PAG)包埋,结果证明按T/C比为3:30时固定化的酶活性明显高于T/C比为8:19者,二者的相对活性分别为58%和6%。用戊二醛将PAL固定于Bio—Gel P—300上的酶活性高于用碘化—1—[环已基—3—(3—二甲氨丙基)]碳二亚胺(CDC)固定化的酶,相对活性分别为63%和18%。
This article reports the preliminary results of preparing immobilized phenylalanine ammonia lyase (PAL). PAL was immobilized using both methods of entrapment and covalent attachment.
