目的探讨不同浓度葡萄糖在不同时间对系膜细胞产生细胞外基质(ECM)的影响。方法人系膜细胞分别在不同浓度D-葡萄糖(5.5、10、20、30mmol/L)条件下培养不同时间(6、12、24、48h),ELISA法检测各组细胞上清液纤维连接蛋白(Fn)、Ⅳ型胶原(ColⅣ)含量。结果(1)10、20、30mmol/LD-葡萄糖条件下,培养6、12、24、48h均能引起细胞上清液中的Fn、ColⅣ显著升高(P<0.05),以24h最明显(P<0.05)。(2)在相同的时间点上,随浓度增加,5.5、10、20、30mmol/LD-葡萄糖培养均能引起细胞上清液中的Fn、ColⅣ显著升高(P<0.05),以30mmol/LD-葡萄糖组最明显(P<0.05)。结论葡萄糖可以时间和剂量依赖的方式刺激系膜细胞Fn、ColⅣ产生。
Objective To investigate the effects of different concentrations of glucose on the extracellular matrix (ECM) production of mesangial cells at different times. Methods Human mesangial cells were cultured in different concentrations of D-glucose (5.5, 10, 20, 30mmol / L) for different time (6,12,24,48 h), ELISA assay was used to detect the cell supernatant fibronectin (Fn), type Ⅳ collagen (Col Ⅳ) content. Results (1) Under the conditions of 10, 20, 30mmol / L-glucose, the Fn and ColⅣ in the supernatant of the cells were significantly increased at 6, 12, 24 and 48h (P <0.05) P <0.05). (2) At the same time point, with the increase of concentration, Fn and ColⅣ of 5.5, 10, 20, 30mmol / LD-glucose could all significantly increase in supernatant (P < LD-glucose group was the most obvious (P <0.05). Conclusion Glucose can stimulate mesangial cells Fn and Col Ⅳ production in a time and dose-dependent manner.
