本研究利用脐血CD34+细胞在体外大量扩增红系细胞为解决血源短缺、杜绝输血传染、克服稀有血型患者疑难配血等问题提供一种有益的途径。利用添加SCF、IL-3、EPO等细胞因子的无血清培养液扩增脐血来源的CD34+细胞,并通过间充质干细胞的支持培养来诱导其向红系细胞扩增和分化。结果表明,经过23天培养以后,本方法可以使细胞的扩增倍数达到2.52×105,其中95%以上都是红系细胞,粒单系细胞和巨核细胞所占比例都在1%以内。无间充质干细胞的培养体系在细胞扩增数量及红系细胞所占比例方面都明显逊于有间充质干细胞支持的培养体系。结论:利用细胞因子和间充质干细胞可以在体外大量扩增红系细胞,其中间充质干细胞对红系细胞增殖和分化起重要的促进作用。
In this study, the use of cord blood CD34 + cells in large numbers of expanded erythroid cells in vitro to solve the shortage of blood supply, to prevent transfusion infection, to overcome the problem of rare blood patients with blood and other issues provide a useful way. Umbilical cord blood-derived CD34 + cells were expanded in serum-free medium supplemented with cytokines such as SCF, IL-3, and EPO and induced to proliferate and differentiate into erythroid cells by supporting culture of mesenchymal stem cells. The results showed that after 23 days of culture, this method can make the cell expansion multiple 2.52 × 105, of which more than 95% are erythroid cells, granulocyte mononuclear cells and megakaryocytes accounted for less than 1%. Mesenchymal stem cell-free culture systems were significantly less invasive than mesenchymal stem cell-supported culture systems in terms of the number of cells expanded and the proportion of erythroid cells. Conclusion: The use of cytokines and mesenchymal stem cells can greatly expand erythroid cells in vitro, including mesenchymal stem cells on erythroid cells proliferation and differentiation play an important role.
