用焦锑酸钾法对心肌内钙进行细胞化学定位,经X射线能量失散谱分析(EDX)法显示,OPP法及PPP法对细胞内结合钙均有特异性反应,OPP法更为敏感。钙细胞化学证明心肌内钙的分布是高度隔室化的,其主要钙贮存池是肌浆网和线粒体,其中肌浆网含钙量更高,焦锑酸钾钙细胞化学定位及EDX能谱分析对进一步探讨钙在心肌细胞代谢及损伤过程中的作用机制有重要意义。
The intracellular calcium was chemically localized by potassium pyroantimonate method. The X-ray energy dispersive spectrum analysis (EDX) showed that the OPP and PPP methods both reacted specifically with intracellular calcium, and the OPP method was more sensitive. Calcium cytochemistry proves that the distribution of intramyocardial calcium is highly compartmentalized and that its main calcium storage reservoir is sarcoplasmic reticulum and mitochondria, with a higher calcium content in sarcoplasmic reticulum, chemical localization of potassium pyroantimonate calcium and EDX energy spectrum Analysis is of great significance to further explore the mechanism of calcium in cardiomyocyte metabolism and injury process.
